Overview
The KrosFlo® TFDF® System uniquely merges tangential flow (TF) with depth filtration to address challenges in viral vector and gene-therapy upstream processing. Its proprietary TFDF® filter captures cells and debris effectively in a robust depth structure, while maintaining high flux thanks to the tangential flow design. This hybrid technology supports high-density cultures including AAV, lentivirus, oncolytic viruses, retrovirus, exosomes, and microcarrier-based cells.
Equipped with closed, gamma-irradiated ProConnex® TFDF® Flow Paths—pre-assembled with disposable tubing, sensors, and pump head—the system provides a ready-to-use configuration, minimizing setup time and contamination risk. The hardware and software are tightly integrated: bi-directional communication enables scale-up from lab to process with consistent control logic, and optional real-time process analytics allows monitoring permeate flow, pressure, and other critical parameters.
With scalable modularity, TFDF systems adapt from lab (1 L) to pilot (up to 500 L) and process-scale volumes (up to 2,000 L). The system uses a minimal footprint (~1 m²) and is designed for linear scale-up by adjusting tube number and length. TFDF technology therefore enables simplified clarification, higher viral yields, and efficient bioprocess intensification for advanced therapies.
